采用牛血清白蛋白偶联的三聚氰胺(melamine-BSA)完全抗原免疫BALB/C小鼠,应用杂交瘤技术制备抗三聚氰胺的特异性单克隆抗体。所得单克隆抗体效价可达5.0×105,且抗体的亚类类型为IgG1,抗体的亲和常数为3.21×109 L/mol。与三聚氰酸、三聚氰酸二酰胺、三聚氰酸一酰胺的交叉反应率分别为30%、93%和46%。说明该单抗可用于三聚氰胺类物质的检测,为研制高质量的三聚氰胺ELISA试剂盒和胶体金试纸条奠定了基础。

英文摘要:

In this study,mice BALB/C were immunized by melamine-BSA and anti-melamine monoclonal antibody(McAb) was prepared by using hybridoma technique.The ELISA titers of ascite was exceeded 50,0000.Subtype of the McAb was IgG1 and the affinity constant was 3.21 109 L/mol.The cross-reactivity with Cyanuric acid,cyanurodiamide and ammelide was 30%,93% and 46% respectively.The monoclonal antibody can be used for fast determination of melamine substances.
