采用快速有效的数学统计方法对堀越氏芽孢杆菌(Bacillus horikoshii)S184产河豚毒素的发酵条件进行了优化。利用Plackett-Burman设计,从众多影响产河豚毒素的因素中筛选出影响较大的3个因素:蛋白胨、磷酸盐质量浓度和接种体积分数。在此基础上,再利用响应面法中的杂合设计进行优化,通过拟合得到响应曲面函数,并获得了最佳的实验条件。在该实验条件下,河豚毒素产量从666.65 ng/L提高到1 900.60 ng/L。

In this manuscript,factorial design and response surface techniques were used to design and optimize the nutrient and environmental conditions for tetrodotoxin production from Bacillus horikoshii S184.The content of peptone and phosphate,inoculum has been identified as three significant parameters for tetrodotoxin production by Plackett-Burman design.Based on those results,the optimum conditions were achieved by response surface techniques.With the optimum conditions,the yield of tetrodotoxin was increased to 1 900.60 ng/L from 666.65 ng/L.
