利用MTT法研究了粘细菌Stigmatella WXNXJ-B代谢产物对HepG2肿瘤细胞的抑制作用,考察了一些理化因素对代谢产物稳定性的影响并定性分析了代谢产物。结果表明,Stigma-tella WXNXJ-B菌株产生的活性物质能抑制肿瘤细胞的生长,抗肿瘤活性物质产生的最佳发酵时间为7 d。大孔树脂HPD100、XAD16和DA201对活性物质的吸附效果较好,该活性物质在12 h的自然光和紫外光照射下、在温度不高于70℃和pH 3~10之间稳定性好,代谢产物中含有内酯类、苷类和甾体类物质。

英文摘要:

In this study,the antitumor effect of the metabolites from Stigmatella WXNXJ-B was analyzing by HepG2 cell using MTT method.Furthermore,the stability of the metabolites under different physical and chemical environment was examined and the qualitative characteristics of the metabolites were analysis.The results showed that the antitumor bioactive metabolites efficient inhibited the growth of HepG2 cell.The optimum fermentation time of antitumor substance was 7 days.HPD100,XAD16 and DA201 resin were efficient to adsorb the bioactive metabolites.The antitumor bioactivity on HepG2 cell line did not obviously change when the temperature was lower 70 ℃,pH was in the range from 3 to 10 and the metabolites were treated with daylight and ultraviolet light during 12 h.It was detected that lactones,glycosides and steroids components existed in the metabolites.
