中文摘要:

通过W/O/W两步乳化技术和乳化-溶剂挥发法制备装载亲水性大分子的固体脂质纳米粒。结果表明:聚乳酸-羟基乙酸共聚物(PLGA)是一种有效的乳化剂,其浓度对粒子的大小、电位有一定影响。乳化工艺为第一步乳化超声20 s和第二步乳化超声30 s,选用二氯甲烷-丙酮作为有机溶剂,乳化剂PLGA的添加量为15%,此时包封率达到58.75%,得到的粒子形态均一、分布均匀;体外释放试验表明:释药性能符合长效制剂特征,且释放后的酶结构并未遭到严重破坏。

英文摘要:

The aim of this manuscript is to prepare the catalase-loaded solid lipid nanoparticles using the W/O/W double emulsion and solvent evaporation techniques.The results showed that PLGA was an efficient emulsifier for the preparation,the concentration of PLGA had an effect on the particle size and zeta potential.From the results we can conclude that the sonication time for the W/O emulsion was 20 s,the sonication time for the W/O/W double emulsion was 30s,Methylene chloride/acetone(1/1) was selected as the organic solvent,the concentration of PLGA was 15%,and the encapsulation efficiency was achieved at 58.75%,also we got the homogeneous particles.The vitro release results showed Higuchi equation could apply to this release curves.This results presented here indicated that the carrier can prolong the action time of the catalase.The CAT structure also was not damaged during this process through determining the activity.
