| 建立了采用超高效液相色谱法(UPLC)测定桦褐孔菌发酵菌丝体中的白桦脂醇、麦角甾醇、羊毛甾醇、胆甾醇、豆甾醇和谷甾醇的质量分数的方法。色谱条件以shim-pack XR-ODSⅢ柱(150 mm×2 mm,2.2 μm)进行分离,流动相为乙腈100%,流速为0.8 mL/min,检测波长为202 nm,柱温为30 ℃。结果表明,UPLC法具有很好的重复性和回收率。甾类化合物分析的日内和日间相对标准偏差分别为0.10%~0.33%(n=5)和0.07%~1.66%(n=5)。获得了在0.3~1.6 μg范围内很好的线性范围。白桦脂醇、麦角甾醇、羊毛甾醇、胆甾醇、豆甾醇和谷甾醇的回收率分为97.05%,98.74%,95.65%,101.45%,96.62%和96.14%。UPLC法适合用来快速、准确定量测定桦褐孔菌发酵菌丝体中6种甾类化合物。

| A ultra performace liquid chromatography(UPLC) method was applied to determinating the contents of betulin,ergosterol,lanosterol,cholesterol,stigmasterol and sitosterol in the submerged-cultured mycelia of Inonotus obliquus. The procedure was carried out on a shim-pack XR-ODSⅢ column(150 mm×2 mm,2.2 μm),using acetonitrile(100%) as mobile phase with a flow rate of 0.8 mL/min. The detecting wavelength was 202 nm and the column temperature was 30 ℃. This method had good reproducibility and satisfactory recoveries The relative standard deviations of this method were less than 0.10%~0.33%(n=5) and 0.07%~1.66%(n=5) for intraday and inter-day assays,respectively. A good linear correlation was obtained in a range of 0.3~1.6 μg. The recoveries of betulin,ergosterol,lanosterol,cholesterol,stigmasterol,and sitosterol were 97.05%,98.74%,95.65%,101.45%,96.62%和96.14%,respectively. This method could be applied to evaluate real samples,and it was rapid,accurate and suitable for the quantitative determination of the six steroids in the submerged-cultured mycelia of Inonotus obliquus.
