从浙江省台州市剑门港海区采集海底淤泥样本,以海带作为唯一营养因子筛选出3株SOD产生菌,并对其中1株产SOD活性较高的菌株从从形态学、生理生化及分子生物学特征等方面进行了鉴定,该菌属于芽胞杆菌科(Bacillaceae),芽胞杆菌属(Bacillus),蜡样芽胞杆菌(cereus),命名为Bacillus cereus MBRH3。利用中心组合设计实验(Contral Composite Design)对该菌株的产酶培养基进行了优化,结果表明,当培养基主要成分玉米浆粉、海带浆、葡萄糖和硫酸锰质量浓度(g/L)分别为4.5、4.11、2.10和0.62时,SOD活性最高达到203.38 U/mL。还未曾发现该菌能够利用海带生产SOD的文献报道。本菌株保藏于中国典型微生物保藏中心(保藏号:CCTCC NO:M2013581)。
英文摘要:
Three Microorganism with SOD production were separated from the seafloor mud of Jianmen harbour in Taizhou, Zhejiang Province, China. Thus, further studying on one of the stronger SOD activity strain was undertaken, Its phenotype, genetic and phylogenetic analysis experiments suggested that the strain belongs to the family Bacillaceae, genus Bacillus and named as Bacillus cereus MBRH3. A sequential statistical methodology comprising of Contral Composite Design were applied to enhance the fermentative production of SOD, As a result, maximum chitinase activity of 203.38 U/mL was obtained in the optimized medium concentration(g/L) of 4.5 corn steep powder, 4.11 Sea weed paste, 2.10 glucose and 0.62 magnesium sulfate. Bacteria was deposited into China Center for Type CultureCollection(CCTCC NO:M2013581).
